چکیده
An efficient encapsulation and slow growth conservation protocol was developed for Nyctanthes arbor-tristis L. an antiviral medicinal plant of the family Oleaceae. A gel matrix with 3% sodium alginate 100 mM calcium chloride (CaCl2⋅2H2O) found best nodal segments. The viability shoot development potential encapsulated segments optimized. Encapsulated stored at 4 °C 24 remained viable up to 90 days showed 42.89 ± 6.04% 33.53 7.15% respectively. Nodal maintained under conditions 180 on one-eighth strength Murashige Skoog (MS) medium supplemented 0.5% sucrose suitable satisfactory (40.28 2.04%), while further addition 0.5 mg/l abscisic acid supported 40.36 1.01% rooting response achieved half-strength MS indole-3-acetic acid. field survival rooted plants 65%. clonal fidelity in-vitro derived plantlets studied start codon targeted primer profile, which same banding mobility patterns as source parent plant. maximum profile monomorphic consistent, confirming stability regenerated plants. method will permit this species facilitate easy exchange germplasm.
